Planapo 63x/1.4, DIC, 90 µm, stacked in Photoshop, Marine sample, Alboran Sea.

top = images 600-642 , bottom = images 635-672 and all = images 600-672

probably Amphora arcus (to be confirmed)

Wow ! These images are amazing. I'm, curious how much time do images like this require ? Assuming one is proficient with the appropriate PS techniques ?

I did not do the whole process in one go but if I had it would have taken a couple of hours.

The photographic session took about an hour from when I spotted the diatom and took the first images to when I went back to it later to see if it had slowed down enough to try again.

I could have done it in less if I had leached the water out rather than let it evaporate, but I was not sure what else was on the slide.

Post processing in Camara raw + photoshop took around an hour or so for the 3 images.

Great images 75RR!

Do you know how big are the steps in your z-stack? I find that 1 µm (or smaller) vertical steps are ok for diatoms, otherwise I get artifacts in the striae stitching.

A little less than one is probably about right.

Had a look just now at the amount of movement I get within the 2µm marks (microscope side on and reproducing the rotation feel) and reckon I get about 3 images per 2µm


On a slightly related issue, I have been disappointed with how little my new batteries seemed to last, until that is I read somewhere that they are supposed to give around 300 shots per charge!

So I stopped moaning, as it's not the battery - it's the fine focus ;)

How many miliampere hours is your new battery and how many is the old one? I hate it when there is some cool microbe or something exciting going on, then the battery dies and I have no replacement.

Folks, are there no AC adapters for Olympus cameras ? for my Canon I bought a (~20-30$ or even less, cannot remember) a 220V AC adapter. A Chinese thing.
And I never touched the battery anymore, now for nearly 2 years.
Had a look at one point but did not find anything for the Olympus Ep-2
