Stauroneis phoenicenteron

#1 Post by Wes » Sun Sep 22, 2019 9:58 pm

A stack of 6 images with total stack depth of 6 µm. 40/0,95 objective DIC.

The diatom is from an 8 form test plate by Klaus Kemp (whom I've not been able to contact recently, I hope he's ok).

Very beautiful photo Wes! Love the depth. And I do hope Klaus Kemp is doing well.

Thank you Timemaster1212 and 75RR. Rotating the stage such that the longitudinal axis of the diatom is at 90 degrees to the DIC axis of shearing improved resolution of the pores quite a bit.

Indeed! Very useful for Pol, DIC and Oblique

Here are some instructions for centering the stage:

On my microscope I need to have two special keys that have square holes with a 2 mm side. I found a substitute that randomly fitted ok and used it to center the stage but it wasn't super straightforward, I had to do a bunch of corrections until it find a workable center.

Phantastic photo Wes.
Thank you Dan and Jochen, I am currently looking into making permanent diatom slides and make more photos like this.
