Re: Substituting LED bulb for incandescent bulb

The wiring'll be fine, you already noted that it will pull half the wattage. There are sometimes issues when filament position is critical, but I don't think it's too sensitive with the optilume.

Re: Good Stereo Microscope?

I like the b&l sz7 but you might want to stay away if you aren't a hands in kind of person as almost all of them need a little work. An sz4 is cheaper and they made a million of them. Sometimes can be had for very little. They're not the most superb imaging available but they are madly convenient. T...

Yeah you oughta talk to Apochronaut about buying direct if you're contemplating laying down six or seven thou on a microscope. A little more legwork maybe but might be worth your time.

Re: Types of Phase Contrast and when to use which type

The Galen phase lenses are not compatible with either the regular B&L phase or the flat field. The regular and flat field phase lenses/ diaphragms are close, except the 10x. The Galen lenses are pretty good looking plan dark medium phase achromat lenses! It is uncommon to see the proper turret, part...

Re: Inverted vs Stereoscope or Compound for Pond water organisms

AN inverted scope is best used to look at things that sink to the bottom so the can press up against the glass, or things you need to access while looking at them. You can get LWD condensers with NA of .5 readily-- they are pretty big and heavy and don't always fit. I use a .4 Nikon condenser when u...

Re: Amateur microscopy and professional micrometer

Yeah, using a big competent microtome the 10 μm setting is for course shaping of the block, then you set it to about half of that to cut the ribbon. Thinner than 5 μm is what you might call "ultramicrotomy," but in any case even your regular microtome should do about an order of magnitude thinner (a...

Re: Epi-Fluorescence Microscopy and Deconvolution

I guess I am confused, does Photoshop have a deconvolution algorithm? I assumed that was what was being discussed as gnu image manipulation program has a genuine one and it is merely a free clone of whatever version Photoshop was at a few years ago.

Re: Epi-Fluorescence Microscopy and Deconvolution

I am not immediately familiar with Photoshop, but I assume when Berdan said he was using Photoshop deconvolution he was using some implementation of a deconvolution algorithm in Photoshop, not one of photoshops non-deconvolution algorithms. I would argue that a method that estimates a psf just from ...

Re: Zeiss GFL compatible objectives?

The epiplan hd objectives require a special set up, larger threads and different lighting. No problem, the others work well. If I'm not mistaken, all Zeiss objectives from that era were designed to work with compensating eyepieces, so they should be interchangeable. The neofluars are famous for.bein...

Re: Epi-Fluorescence Microscopy and Deconvolution

Call me a numbskull, but I don't understand how you'd deconvolve an image without having a solid grip on how the image was convoluted in the first place. I guess that why you'd (have to) figure out the point spread function of your optical system and then feed that into a specifically designed de-c...

Also, if you get a chance to read the paper I think you will find that a "poor man's Kohler" is exactly Kohler, as he had limited equipment but a good understanding of the principles and a knack for ingenious improvisation.

Re: Poor man’s Kohler illumination

You can find and read Kohler's paper--its actually quite entertaining and not really overly technical. Although he did make use of a field aperture, really the essential components are a field lens and an adjustable condenser. Without these you can't really get the even illumination effect. Without ...

Kohler himself used cardboard with holes cut in them. It seems also like you could focus on the frosted glass and then simply move the condenser up about 10mm or so.

Re: What are the most common inverted scopes?

Bausch and Lomb presented an inverted scope in the 19th century. It didn't really catch on right away and they stopped making inverted scopes until the projection scope design from mid century or the I think Photozoom a little while later. They also sold a few inverted metalloscopes now and then tha...
