"baby" physarum polycephalum from spores
Hello, 3 weeks ago I put spores of physarum on agar gelose 1% (with no nutrive elements) with a thin layer of water. I could see the flagelate and amiboid forms for days... a few days ago i added 1 or 2 oat flakes... and today I could see the "newly born" physarum : they are tiny plasmode form for t...

I'm living in France.. so i dont know about the "Lochoptera dubia" but i'll ask the entomology dept at work. And about the carcinogeny of acridine orange can you give any data and sources ? And aceto -orcein is hard to buy and also pretty expensive. The amount needed of acridin orange was really rea...

So I tried with acetic acid 100% alone and then I rince and dye with gentian violet (crystal violet). It works but it shows more blurring of the definition... 21 juin 21 chromosome chironome fixation acétique violet x20 2.jpg 21 juin 21 chromosome chironome fixation acétique violet x20 3.jpg 11 juin...

In fact I've just found out after a fex trial the missing important information : None of the protocols mention FIXATION and i'm going to show you the effect of the addition of a few drops of a mix of alcool-acetique acide : its immediat : the DNA precipitate and the chromosomes become visibles even...

Hello, I 've found out that there was a protocol to observe the giant chromosomes présent inside the nucleus of the salivary glands of blood worms ( larvae of chironomes). I wasnt successfull in dyeing them properly yet with methylen blue or toluidin blue so I tried with orange acridine under Uv-blu...

Re: First try at Fluorescence!
this is really pretty good for a first but I think you should try a filter of a bit deeper yellow to get rid of most of blue light and enhance the fluorescence.
Try with some moss or pond alguae as they fluoresce in bright red

Hello, I've just received my photo eyepiece Olympus NKF x3.3 to use with my BH2 :) Indeed the optical quality is much better !!! But the best thing is when I tried it with my Nikon D3100 instead of my astr cam ZWO ASI 120MC that I usually use. The color management is much much better and for once I ...

Re: Physarum polycephalum : "blood" circulation
Hello Sir, thank you very much in fact I have no protocol and I just put the petri dish without its lid on the stage and used the X10 objective. But you can do the same in bright field . In fact the easiest way would be to make an agar plate in a smal petri dish but with a very thin layer eg 1mm thi...

Physarum polycephalum : "blood" circulation
Hello, I have several sample of the "Blob" physarum polycephalum in petri dishes and as i'm preparing sclerotium (dried dormant form) i decided to put some pseudopodes under the microscope. But the thing is that in bright field you dont see much as the creature is quite thick so I thought that maybe...

Re: DIY Fluorescence Microscopy
as a fluorescent dye I mainly use the orange acridin (you can buy it on ebay at a really good price) as it changes color depending on the pH and if it links to DNA or RNA ; dead or alive matter... It works great with live protozoa and bacteriae for example.. 3W led @ 455nm or 470nm are available on ...
