Re: compare bright field and phase contrast
Thanks Greg!
And, after looking at it again, I believe my initial ID was incorrect. I would say the algae is Monactinus judging by the cell shapes.

still figuring out the phase contrast kit from Amscope - here are a couple of images of what I believe to be Sorastrum from a sample from the Saint Lucie River, taken with Amscope's MU503 camera on the T660 fitted with their PCT200 phase contrast turret and 40x plan ph objective: PhaseContrastCompBF...

Finally got the correct phase contrast turret for my Amscope T660. Just quickly experimenting...here are a couple of shots with the 40x. I think I may like this

Re: AMSCOPE 660 TURRET
I just received (Saturday) the replacement for the phase turret for my 660 that Amscope had originally sent that wouldn't fit the dovetail on the condenser holder. The new one fits and works as Greg says - I didn't notice a problem with the brightfield iris (I'll check later today).

I also think these are very good images. Nice work! Btw what work do you need done on your Phomi III? Thanks Wes! As for the Photomic, it needs some lubrication and cleaning (easy enough), a replacement bulb (ordered), just received the 47-3024 photo tube (to use with a Nikon DSLR), and figuring ou...

Thank you, Greg!
Glad you only dissolved the lens coating and didn't delaminate the lens itself. I've been pretty happy with the 660, a good value within its limits.

Could it be a grain of pollen? hmmm...hadn't thought of that. Thank you! After all the desmids and other green algae and cyanobacteria I was seeing in the sample...maybe even a sporangium of some kind, I'll have to see if I can find some more and what is flowering (most likely a Utricularia, I didn...

Re: from the Savannas Preserve
Thank you, 75RR! These are all brightfield illumination of wet (live) mount slides. The Amscope T660c microscope does allow for Kohler illumination, which definitely helps. The camera is Amscope's MU503 via ToupView. For the stacked images, I use Zerene Stacker. Finally, I usually set the white poin...
