Re: Home built laser scanning confocal microscope
Now, i am focusing on scanning head modeling. After the system finished, i will test the confocal imaging quality between different photomultiplier tubes, and try to use SiPM module designed by Vincent et al. as detector. Very interesting! Here are some more links that might be relevant for the pro...

Re: Home built laser scanning confocal microscope
The commercial confocal microscope is tooooooooo expensive for personal use, so, i try to make it in low cost using used parts.(Just for prototype~) :D :D :D Do you already have an idea which galvoscanner and which photomultiplier tube you would like to use? If you haven't seen yet, Thorlabs offers...

Re: Home built laser scanning confocal microscope
Next, i want to design my own scanning head, and hope better than this. :D :D :D I would be glad to hear more about that when you progress with this project! I used custom LabView programm to control the system. The objectives is olympus PLN20X, the tube is removed from olymupus cx21 binocular obse...

Re: Spammers proliferate
They are taking a break and counting shoes. Seems there are plenty of shoes left, and spammers take over the forum once more. This is a nuisance for the users as well as for the administrator of the forum. I do not understand why the offer of long-standing contributors to help solving this problem ...

Re: Olympus and Leica
I'm thinking of two possibilities: Olympus BX50 or Leica DMLB. Both are of much more recent vintage than an Orthoplan or a Universal and both are upgradeable to PC and even DIC. Anyone have advice on the relative merits of these two systems? I use a Leica DM 2000, which is quite similar to the DMLB...

Great work, thanks for showing!
Can you give some more details on the software you used for controlling the scanner and on the utilized objectives and tube as well as scanning lenses?

Re: DSLR vs dedicated microscope camera ?
I agree with everything that has been said above. My personal opinion: I would only use a DSLR if you already own it. If you buy a camera, I would prefer mirrorless in case your focus is on obtaining colorful and crisp photomicrographs. If you focus is on documenting and image processing (measuring ...

Re: Mirrorless camera and adapter combination?
Your setup looks very good! In addition to the 0,5x lens, many manufacturers also provide 0,63x lenses. But in your case, the setup without any lens seems to work very well, why change it? Overall you have a very compact and usable system, congratulation! The E-M1 camera will deliver high-quality im...

Re: Mirrorless camera and adapter combination?
So doing the math and assuming that 12mp is far more than enough while using 10x or 40x objective on my microscopy...and with mainly wanting to do live viewing on a computer with some pictures taken and video recorded in HD. Would there be any advantage of getting something like: Olympus OM-D E-M1 ...

Re: Mirrorless camera and adapter combination?
I can mount the camera body (without lens) directly to the C mount if I have the correct adapter piece, correct? Yes, that's correct. This is one of the big benefits when using infinity optics: The objective projects an image to infinity. The tube lens corrects (almost) all lens errors of the objec...

Re: Mirrorless camera and adapter combination?
Hi cwilli, first welcome to the forum! Your microscope is a setup with infinity optics, which makes camera adaptations rather simple and straightforward. It's certainly better to attach the camera to the trinocular port. From the manual it seems that your trinocular head is equipped with a c-mount, ...

Re: Who's Using Fluorescent Markers and Tags?
There are many textbooks about fluorescence microscopy, but none of them targets "idiots". This is certainly true, since fluorescence microscopy requires quite some technical skills, and also caution when dealing with the frequently toxic and/or carcinogenic fluorophores. The recently published boo...

@Louise, Hobbyst46, FatBassPlayer Alexander: thanks for your encouraging feedback! The stem actually turned out quite well. I also had a look at the leaves, searching for the glandular hairs and the trichomes. Making thin (<20µm) section sof the leaves turned out to be a difficult task. But finally ...

Re: Meadow sage under the microscope
I used the stem of one plant to watch it under the microscope. For this purpose, I fixed the stem in a mix of ethanol, formalin and acetic acid (AFA). Afer 24hrs, the AFA was replaced by 70% ethanol, followed by embedding in PEG (12hrs in water/20%PEG, 12hrs in water/50% PEG, and finally 12hrs in pu...

Meadow sage under the microscope
The meadow sage ( Salvia pratensis ) is a pretty common plant in Europe, Asia and northern Africa. It likes sunny, non-fertilized meadows and typically blooms from end of May into June. As can be seen from Fig. 1, the plant can be quite abundant in some places. The picture was taken in the south of ...

Thanks for the feedback! If I have something to complain, it is the missing scale bar. Without that, I never know how large these animals are. Re: Leica Fan here
For the 100X oil, I might explore that avenue in the future. But do note that, in order to have the best working environment for that objective, you'd have to also replace the dry condenser with an oil condenser. Then I'd want DIC for that, too, that would require a K4 condenser prism and a DIC pri...
