Re: How do you deal with the glare in camera

A couple of thoughts: Check your kohler alignment and make sure the field stop is opened sufficiently. This looks like a "hot spot" from a mis-aligned condenser. Kohler alignment can change with cameras, as the adaptors (or lack thereof) that you add to the optical path can change the focal length o...

Re: Laminar flow equipment

Your best bet for DIY option would be one of the HEPA laminar flow systems mushroom growers use. These produce a continuous stream of sterile air that you can work in. Downside is they only protect your samples (and actually blow your samples into your face), so you have to be cautious of what you a...

Re: Do we always know when a microbe is eating?

Depends on the microbe. Bacteria generally acquire nutrients by secreting enzymes that break down food in the environment which is then imported by specialized proteins into the bacteria. Some don't even both with the enzymes and just import what they can. Most algae photosynthesize and make their o...

MRS is the standard bacteriological medium used to grow lactic acid bacteria, and will nicely support the grow all three species in your probiotic capsules. However, Bifidobacterium longum is a strict anaerobe and is unlikely to grow under the conditions that you would have in a home lab. Both Strep...

You shouldn't have to recalibrate every time, so long as your camera mounts in more-or-less the same place each time.

Re: Water Sample Disposal

Down the sink is fine - keep in mind, every time you flush your toilet after going "number 2" that you are putting a massive bolus of bacteria - many of them pathogenic - into the sewage system. The organisms in your pond water samples are in minute numbers/concretions compared to even the smallest ...

Re: What is a tumor?

People have gallstones. What are they? Your gall bladder makes bile acids, which are chemicals that help digest your food. The bile acids normally pass through ducts in the gall bladder into the small intestine. Sometimes the bile acids can crystalize, forming a stone-like mass that blocks the bile...

Re: What is a tumor?

Very interesting. You are saying the disease process in plants is parallel and similar to cancer in animals. There is a difference in the way and extent the dysfunction spreads, but that is pretty technical. Is "cancer" generally recognized as a technical or scientific term, or just common usage? C...

Re: What is a tumor?

The argument is firmly put to close here: "Like animal cells, plant cells mutate, but does that mean that they can develop cancer? In fact plants may not get cancer as we think of it, but they can and do suffer from tumours, where cells become disorganised and divide uncontrollably." But I don't se...

Steel shims for permanent slides

I recently received a prepared slide (containing a cross-section of a stone pine needle) that we are using as a standard while trialing some new scopes and analysis software. The slide is prepared in an interesting way - a thin steel ring was glued to a slide to create a well into which the sample w...

Re: The floating coverslip

That is an amazing technique. Reminds me of the "hanging drop" method we use in the lab to reduce the amount of antibody (which is stupidly expensive) we use to immunolable samples. A small amount of the antibody solution is placed on a hydrophobic surface, and the sample on a coverslip "floated" on...

Re: measuring psf without fluorescence

Thanks for mentioning wavelength-dependency, I was also wondering this. So it actually makes a lot more sense for fluorescence than for (white-)light microscopy. I already tried psfgenerator and deconvolutionlab2. There still is a PSF with white light, its just that it is not as defined as with flu...

Re: measuring psf without fluorescence

PSF's are wavelength-dependent, so unless you're imaging with light through a fairly tight notch filter, you're probably not going to be able to measure a PSF very effectively as each wavelength of light passing through your subject will create its own PSF, which will overlap with the PSF of the oth...

Re: Stupid new comer question

I teach microscopy to undergrad students, and your confusion as to how magnification works with digital cameras is completely normal. Through the eye pieces it makes sense - something at 100X magnification will look 100X larger than in normal life. But with digital images, you can zoom in and out, l...

There are a number of 3D printed microtomes out there - most variations on the "spindle and bolt" hand-made versions. This is similar, but is a much better system than any other I've seen - as soon as I fix my printer I'm going to give it a try!

Thanks for the ideas everyone. I was able to crack through the ice on a small creek on my property and pull out a bit of algae. Seems to be some diatoms and cilliates in the sample, which will be good enough for today. I really need to get an aquarium going!

I have been asked to bring a microscopy sample to an event - tomorrow - but have an issue. I'm in Canada, it's still winter, and my two best source (a pond and birdbath) are frozen solid. I found this article while looking for something else. Turns out its rather easy to raise your own tardigrades (water bears, moss piglets, or whatever else you want to call them), needing nothing more than clean water and a source of algae. Looks like a fun project! https://www.tandfonline.com/doi/f...
