Hi Mintaka,
Sorry misread post. If flipped 90 degrees, (rather than the 180 I thought it was), that would be one Girdle view and one Plan view of the diatom.

Re: Rocking microtome practice session
Hi Naphthalene, What are the samples you are processing currently? And what are their (approximate) dimensions? Maybe I'm wrong but It appears to me that your samples are kinda too large. In my experience I used samples below 1x1 cm in length/width, but I leave open the possibility that larger sampl...

Re: Leeuwenhoek Microscope Copies
Thanks for this. Interesting. Here is a couple of Leeuwenhoek replica microsopes. I can actually see things with both but the brass one gives the best images.
Looked up how they work and came across a site that tells you how to make one.

Hi Naphthalene,
my Microtome has not arrived yet -
so can only mention that the manual says 20mm by 20mm (actually it says "cuts specimens up to 20mm diameter" see page 3.

Re: A small microorganism
I relied on the calibration marks on the fine focus knob; I guess I should've said "approximately 1 µm" :) Got me wondering what the fine focus graduations represented on my Zeiss RA. Bit fiddly - but basically drew a line (in pencil) across the base/focus block and turned the dial 360˚as it is so ...

Re: Seeking Learned Opinions...
Do you mean to try to substitute the "knife" for a razor blade held by a aluminum bar? Is this in a Cambridge Rocking Microtome? Here is the manual in case you have not download it: http://goo.gl/fkwpCJ The photo at the end of one of the threads showed a DIY (?) razor blade holder that seems as thou...

Re: P.A.C.O filter system and condenser - problem?
My filter was originally below the lens, easier access that way. I swopped them hoping to get better results with darkfield stops by moving it closer to the condenser diaphragm. There is a screw at the bottom of the post which is a little hard to get to, an "L" screwdriver will work best. In the mea...

At 10µm that is very impressive.
How are you getting 1µm focus steps?[Edit] I forgot to say that the first image is a focus stack of 15 images taken at 1 µm focus steps and stacked using CombineZP.

That was a bountiful experiment.
I take it that you used "fresh seawater" as opposed to the reconstituted stuff some aquariumists use?
Fun GIF at the end!

Nice Diatom. Like the brightfield (black and white) image most.
Are there several smaller diatoms under it?

Hi IanW,
I think it would be better if you linked to Jacek's photographs rather than download them and re-post.
You can contact him at: http://www.photomacrography.net/
I am sure he would be happy to tell you what equipment he is using.

My apologies, I should have started out by saying welcome. Welcome!
What equipment do you have? When you have a moment post some images of it in the My microscope section.

Hi Jaguarundi, that is a lot of interesting stuff, emphasis on "a lot". I am a little overwhelmed by the speed you move the slide about as you look for more "interesting" things. Most of what you show could well also be interesting if you gave us a chance to see it. Slow down a little. Will continue...

That is a good space saving idea. Those are the actual microtome's dimensions - it would be easy to simply wrap a big old bath towel around it and carry it in a rucksack or canvas bag say.
Many Thanks.

Just a few problems with that.Go on - you must - don't leave me to blunder about alone!
1) Blew my budget on this already.
2) I am not in Darkest Peru - but I am not far from there!
3) It weighs 10 + Kg That means serious postal charges!
