Sorry misread post. If flipped 90 degrees, (rather than the 180 I thought it was), that would be one Girdle view and one Plan view of the diatom.

Here is a couple of Leeuwenhoek replica microsopes. I can actually see things with both but the brass one gives the best images.

my Microtome has not arrived yet -
so can only mention that the manual says 20mm by 20mm (actually it says "cuts specimens up to 20mm diameter" see page 3.

I relied on the calibration marks on the fine focus knob; I guess I should've said "approximately 1 µm" :) Got me wondering what the fine focus graduations represented on my Zeiss RA. Bit fiddly - but basically drew a line (in pencil) across the base/focus block and turned the dial 360˚as it is so ...

My filter was originally below the lens, easier access that way. I swopped them hoping to get better results with darkfield stops by moving it closer to the condenser diaphragm. At 10µm that is very impressive.
[Edit] I forgot to say that the first image is a focus stack of 15 images taken at 1 µm focus steps and stacked using CombineZP.
How are you getting 1µm focus steps?

That was a bountiful experiment.

Those are the actual microtome's dimensions - it would be easy to simply wrap a big old bath towel around it and carry it in a rucksack or canvas bag say.

Just a few problems with that.

1) Blew my budget on this already.
2) I am not in Darkest Peru - but I am not far from there!
3) It weighs 10 + Kg That means serious postal charges!
