Re: Amphora optical sections
Hi Adam, Thank you for your comment. The diatom measures at 40x60 µm. I used DIC with a 40/0,95 apochromat Zeiss Jena objective, the microscope was setup for afocal projection with a 10/18 Kpl-W eyepiece (with a 40 mm pancake lens on a Canon 50D). The diatom was attached to the underside of the cov...

Amphora optical sections
I would like to share a small series of optical z-sections of an Amphora diatom that had attached to the underside of a floating coverslip. I decided not to stack as in this series of sections you can see different organelles such as the nucleus and chloroplasts in positions relative to each other. ...

Quite a nice finding and superb documentation. Btw you can directly embed the video here in the thread by placing the video ID within the youtube tags
e.g. the ID of your video is HaGDw2e6cBg and you put it [*youtube ]*here*[/youtube*] (without the asterisks)

I can recommend acridine orange, as previously mentioned its very cheap and emits in multiple parts of spectrum depending on local conditions such as pH. Intriguing finding and very well documented at that. I am more inclined to think these are predatory/parasitic bacteria. Maybe Vampirococcus?

One of the best microscopy-related investments I made
was to buy an air purifier. Basically a high grade HEPA filter with a fan that sucks in air and releases completely dust-free air back into your microscopy dedicated area. Not having to deal with dust getting on and in the microscope has been such an improvement and I highly recommend to anyone stru...

It makes you wonder if at some point the bots will get so good they actually start providing useful contributions! Maybe they are already doing that... Have you seen what some of the AI is capable of today? Sometimes I legitimately wonder what % of online interactions are with real people.

Very nice finding Javier. Can you tell me more about its habitat, what kind of pond was it, did you scrape any pond scum, slime, decaying plant matter or similar? Did you take the sample from a shady place or a sunlit area? I have never seen Lacrymaria in the wild. I cannot say I'm happy with the qu...

Re: These 'New Members'
I had no idea but I have seen it on other forums. Maybe the question is too easy? The problem is that the tech is getting better; it's pretty trivial these days to programatically parse a simple question and get the answer. At the point you start tripping up the program, humans tend to be failing a...

Re: Coleps feasting on a rotifer egg
Thank you Tim and Adam Great stuff Wes. The 25x Planapo is my favourite objective - do you have the dedicated DIC slider or are you using a different one? I also like it a lot, it provides a great compromise between magnification, resolution and spherical aberration. I use the slider for the 6,3x pl...

Coleps feasting on a rotifer egg
Coleps is a genus of predatory ciliates that feed on bacteria, algae, other protists (dead and alive) as well as animal and plant tissues. Here you can see one Coleps specimen feeding on what I believe is a rotifer egg. If you look closely you can follow the path of egg particles making their way to...

This microscope comes with an achromat-aplanatic BF-Ph-DF condenser type V Z. So to answer your question, no the condenser does not have DIC function (only phase and darkfield). Cosmetically the microscope looks very nice, and assuming internally there is no delamination and other optics issue this ...
