Help needed with acridine orange.

Hi gang. I would appreciate some instruction for preparing acridine orange. I don't know the best way. Guidance appreciated.

I assume you want to use it for fluorescence microscopy and by preparing I would assume you ask how to make solution and use it instead of synthesizing it yourself. If you have access to DMSO measure out a given amount and prepare a 10mg/ml solution based on the pre-measured mass. Otherwise you can use water and prepare 0.1-1mg/ml. Working concentration would be anywhere between 1-10 µg/ml so use simple volumetric dilution (c1v1=c2v2 where c-concentration, v-volume).

I agree with @Wes. Make a basic solution of 1 g in 100 ml distilled water. From that make a working solution 1 ml basic solution with 99 ml distilled water. For vital cells you must dilute it even more.

Please remember that acridine orange like most fluorescence dyes is a mutagen. While handling the powder, wear breath protection and gloves. Avoid any skin contact and any pollution of your work space. The solution is less dangerous but is to be handled with care as well.
