I am looking for a way to make a permanent "wet mount" of a pond water sample, but I've failed to find a method to do so. The sample will contain a mixture of filamentous algae, diatoms, protists, and bacteria. Would fixing by adding 4% PFA, then allowing the sample to dry to the slide before adding a coverslip and mounting media work?

I have some nematodes mounted in glycerol that are sixty years old or so and still show good detail. Try looking for Seinhorst's method.
