Great thread Red-Green. Thanks. Your pictures got better and better - and the examples provide others with a good idea of what phase contrast adds for near-transparent specimens.

I believe the Mounting size of condenser is 37mm in diameter / bottom mount.

The only cons that I have really found with this phase kit is that the phase plates on the turret sometimes become unaligned especially when switching between darkfield / brightfield and that each one is just adjusted with your fingers. So every so often I have to realign. Not really a big deal, just a slight annoyance.

Other that, it's a pretty amazing kit. The pictures and view that you get from it is outstanding.

IMHO, the PC kit effect is sorry for the price, and the effect produced by one finger (oblique lighting) is enough to surpass it.

Hi Red_green,

In post #13, the olympus 40x picture shows too oblique illumination. Look at the upper left part of the picture. Bacteria
appear with too much 3d effect. This may be a illumination issue, or light and optics alignment problem which can destroy
the phase contrast. For my opinion, this picture does not seem to have a correct phase contrast setup or something
is wrong.

In my job, I use phase contrast daily for live cells using equipment from different brands (olympus of course and nikon and leica as well) and the result is clearly better than the one showed by the mentioned olympus 40x picture. I don’t know what equipment
is used in this picture and may be interesting to know.

On the other hand, the results obtained with your Amscope kit are very good and for all those for whom DIC is out of they budget,
phase contrast will be a new world to discover.

Is there different type of phase kits available? The one I have only produce gray background at the most, not dark and not at this level of contrast. There is no halo around any subject. Yes, it is centered correctly using phase telescope. It is a positive phase objectives (PHP written).

Some of the images in this post are excellent.
