Zeiss Photomicroscope DIC Inko questions.

#1 Post by Seb28 » Mon Apr 05, 2021 2:59 pm

Hi
I am trying to achieve the DIC in my Zeiss Photomicroscope. But I am not sure if I have the correct DIC/Inko condenser.

I use it with the Zeiss DIC slider no.II.

The interferential bands do not match.
The microscope is equipped with a fluorescent module and I wonder if dic works with this fluorescent module or should I remove it and use standard nose piece for objectives?

Hi,
here is my description of the fitting components and how to set them up: https://www.google.com/url?sa=t&rct=j&q ... RH8tTHdZix

I think this kind of DIC depends on specific objectives (e.g. Plan 40/0.65) and it only works within particular serial number brackets of those particular objectives.

Definitely try to regular nosepiece turret if you have it handy.

What is required beside your #2 DIC slide and DIC condenser is the polarizer and analyzer to obtain cross polars first. The #2 DIC slide has the analyzer and you would need a polarizer under the condenser. The equipment should work even with the IIIRS fluoro unit as long as you're not going through fluoro filters in the unit. Do you have the attached instruction? This for the #3 DIC slide for use on the small Standards but the principles are the same. If you want it, send me a PM with your email and I will send it to you.

That's not good news for me.I already ordered the nosepiece turrent.

I know how to use all the DIC components.I tried with Zeiss Neofluar 16x and 40x Ph objectives,Nikon 10x ,20xCFN and Pzo 10x.Best results are on zeiss objectives.
I'm close to the real DIC but something is wrong and I don't know what.

Phase objectives rarely work very well with DIC.

With that fluorescence condenser: Didn't you have to lower the stage? This would disturb Köhler illumination setup. And the fluorescence condenser, does it have a tube factor?
I would suggest to start with one of the tested setups, right objective in the right serial number range, set up as I described, normal nosepiece, not fluorescence or wide field nosepice. From there you con look what other setups give you the right DIC contrast. Some other setups work to a certain degree, very few perfectly and many not at all.

I'd be interested to see some examples of the level of DIC that you achieve with your system.
