algae and protozoa live in UV

#1 Post by chrisimbee » Tue May 04, 2021 6:36 am

hello,

last night I tried to take better pictures with my olympus epifluorescence module :

I dont have the lamp house that goes with it so I have to use different kind of light sources. So far the best ones are a little UV torche (wich is in fact @405nm) or a white light led torch (quite high intensity but i dont know the lumen value).
The module was given to me with a "cube" that selects blue light ( the curve shows 350 to 500 nm). I also tried with a strong led growlight (3500K) used to grow plants but it doesnt work. A halogen lamp doesnt work either , nor does it with a standard led lamp for a desk. But it works fine with both torch and I think mainly because they emmit a large amoung of violet and blue light and foremost because of the presence of a focusing lens in the torch.

So I tried again with some green stuff I found in my cultures, and also with some protists addes with a really tiny amount of rhodamine G.
Rhodamine G at such a low concentration doesnt kill the protozoa immediatly and I had some hours of observation until they die. Rhodamin emmits light at different wavelenght : from green to yellow orange. Here are some of the results that are pretty encouraging. The green algae autofluoresce with a strong red color .

As the light source used isnt strong enough its difficult to directly see the fluorescence at the eyepiece but when the torch is in the right position it works fine and its an incredible view. I have to buy a much stronger source to get better image.
