melosira + pleurosigma

#1 Post by 75RR » Fri May 07, 2021 9:56 pm

.
Zeiss Planapo 63x/1.4, DIC, 20µm width (melosira), stacked in Photoshop, Marine sample, Alboran Sea.

I prefer it when they are on their own, this is messy, still, thought it worth a stack. In this case 54 images.

Lovely! I'm intrigued to know how you manage to stack live subjects - I mean, aren't things moving around a lot?

Thanks LouiseScot

I pin them with the cover slip. To do this i wick the water from under the cover slip using absorbent material - usually tissue paper.

This has two functions, it holds the subject down and places it in the 'sweet spot' re imaging.*

This does not always work of course, even the smallest piece of detritus can stop the cover slip from pinning the subject.

One can also move things around in hopes of getting a better arrangement by adding water and then removing it.

One adds water by placing a drop on the edge of the cover slip - capillary action does the rest.

*(The layer of water between the subject and the bottom of the cover slip effectively adds to the cover slip thickness.)

Count me in as thoroughly admiring that shot.

To me, capturing two side by side like that is 4x the challenge and 4x the excellence when done so well -- compared to equally excellent individual shots.
