Alyssum Flower Stigma Focus Stacked

#1 Post by Sphaerotilus » Sat Jun 05, 2021 6:40 pm

Been playing around with focus stacking photomicroscopy and evaluating a focus stacking program called Helicon Focus.

I believe that the small yellow structure shown here is the Stigma end of the Pistal. My image was created using a Nikon light microscope. Several images focused at different planes were photostacked together using Helicon Focus, with additional post processing work in Lightroom and Photoshop. The polar width of the small granules (pollen?) in this image is approximately 20 microns. You can also see many of the individual yellowish retangular shaped cells making up the Pistal.

My initial impressions of Helicon Focus is great. I find that focus stacking in Photoshop not to be good for microscopy.
