Success - Permanent Mounts

Specimens were set in GALA 40.

GALA 20 was too weak and GALA 60 destroyed everything. So I did a 50/50 mix to create GALA 40.
Left the specimen in there for a few minutes and did a distilled water bath in three successive test tubes. Took out drop after drop on a slide til I found it and sealed the edges of the cover glass with clear nail polish. The slide is about 48 hours old. I think I might have actually done it right. All by intuition and accident.

So, do you mean when you say "left it in there" that you left it in the GALA 40 then water-rinsed? What did you mount it in, just water?
Did you find it with the dropper when it was in a water-bath, then drop the specimen (in a water-bath drop) onto the slide then seal?
What happened with the G-40? I'm having trouble understanding what you actually did - sorry to be so dumb... :oops:

After "setting" the specimen in Gala 40, I removed all that alcohol with a fresh water rinse, then sealed it wet. In the mean time I have another that I have left open to see what happens if you let it dry. I have mounted the specimens in the gala and notice they deteriorate rapidly as the alcohol does it's thing. Experimenting is fun.
We'll check it out again in another 24 hours.

You want to dehydrate the specimen slowly by shifting it to tubes with a successively higher concentration of alcohol. Could take several days for a sensitive subject

Right. The specimen is another daphnia. I'm about to find out how tough it is. 😅

The result of allowing things to completely dry is that diatoms do well. Everything else is gone.
