Amphora optical sections

I would like to share a small series of optical z-sections of an Amphora diatom that had attached to the underside of a floating coverslip. I decided not to stack as in this series of sections you can see different organelles such as the nucleus and chloroplasts in positions relative to each other. From top to bottom the images go from the middle to the top of the diatom.

Great results Wes.

I've recently been experimenting with pushing the resolution limits of my Zeiss Universal on similar subjects, so any technical details would be appreciated. And how big is this diatom?

I missed this one, Wess. This is wonderfull, so beautiful and detailed. I would love to see this stacked too though...

Could you please explain me what is a floating coverslip? You mentioned that on another thread.

You can see the stacked image includes random elements from the diatom and doesn't give you a good idea of how it looks in reality (but still looks interesting).

Oh, I see it now. It looks great, but the layered sequence is so much accurate and detailed.

Thanks for the info Wes. There is a picture of my Universal here: viewtopic.php?p=107684#p107684 but I should really update the thread following the Universal upgrades. The short version is that I don't use my Standard any more, but I use the Universal almost every day!

Are the Zeiss Jena apos plug-and-play compatible with the Zeiss west? 45mm and 160mm? I recall looking at one but then saw it was 35mm.

Stacked images can sometimes look like a object is squashed or compressed, suppressing the visual impression of it's true depth.
Those are very well done slices. Sometimes those diatoms can look like a ship in a bottle.

The Jena objective I used is fully compatible with my Zeiss West scope (photo of this particular lens posted below). I bought a DIC slider for a 40/0,75 water-achromat and by chance it produced really homogeneous DIC with this Jena apo (which, however, is not the case with the Zeiss West 40/0,95 planapo).
