Hard to Get Clear Image of Fibers

Hi, for my research purpose I need to take clear images of single fibers in a yarn under a certain zooming ratio. I purchased Amscope MU1803 18MP camera and attached it to Hayear zoom c-mount lens to capture the images. However when I tried to take images of the yarn, it is always blur and seems hard to get a clear image with single fiber resolved when reached the relatively best focus (others will be just blurry) . I would like to know if there are ways to solve this or there is a better approach to get a sharp image of fibers. Also previously I used a Nikon 745T trinocular microscope but the image quality is also not very good when taking images of fibers.

I'm not an expert, but I think you are facing a depth of field issue. If your lens has an iris, you could try closing it to the point your depth of field and contrast increase, but you lose no resolution. Beyond that, you will need to stack images:

alternatively you put a piece of glass over it and flatten it

Thanks for the advice! However since the yarns is twisted it would be better if it is not flattened for my purpose. For the lens mine doesn't have iris, does getting a iris diaphragm and attaching it to the objective help?

I think a macro photographer has to chime in here.

My thoughts: an iris diaphragm has to be placed in a very specific place of the light path to be optimum. You could try stopping down the aperture of your lens with a simple aperture mask in front of it. You will end up with a lower resolution lens, but it will display more depth of field. It might serve your purposes.

Stacking should certainly help, but … to be brutally honest, I couldn’t see any plane of your image that is sharp.

I wonder if you have a vibration problem ?

You can surely try an iris, cut a piece of cardboard with an hole like 2mm diameter, and stick it in front of the lens, see if improves.

Also, don't take photos at the max magnification, zoom out a bit, like double distance, should get cleaner, more depth of focus (maybe with same amount of detail)
Those zoom lenses don't work well at max magnification.

How large is the diameter of the yarn? 0.5mm? 3mm?

What magnification (or rather field of view) do you need? I agree this looks more like a vibration issie, though if thr hayear camera is cropping the image heavily it could be an optical issue I guess.

Yes it is like that. I am trying to solve the vibration issue since the current stand I am using is not heavy enough.

This setup can provide a 0.7x to 9x magnification and the image is taken under 5x. For the camera I am not cropping the image.
